Cell-free synthesis and processing of the proteins of poliovirus.

چکیده
Poliovirus RNA can be translated completely and accurately in rabbit reticulocyte lysates; the nascent poly-protein is processed to give primary products 1a, X, and 1b indistinguishable from those made in poliovirus-infected HeLa cells. The capsid precursor protein 1a is processed to form the capsid proteins VP0, VP1, and VP3, while the noncapsid precursor 1b is processed to form protein 2.

Identification of RD and L20B Cell Lines Cross-Contamination by Multiplex SYBR-Green PCR in Poliovirus Laboratory
Background and Aims: World Health Organization Global Polio Laboratory Network (GPLN) plays a critical role in the Global Polio Eradication Initiative. Cell culture methods (mostly RD and L20B cell lines) have been used for Enteroviruses and polioviruses isolation, respectively. Cross-contamination among L20B and RD cell lines causes the problem in accuracy of poliovirus surveillance and decrea...
متن کاملComplete translation of poliovirus RNA in a eukaryotic cell-free system.
Poliovirus RNA stimulates imcorporation of 35S from both [35S]methionine and formyl-[35S]methionyl-tRNAfMet in cell-free systems derived from HeLa cells or from poliovirus-infected HeLa cells. The largest product formed under the direction of the viral RNA is the same size as the polyprotein thought to represent translation of the entire RNA. Synthesis of this polyprotein and other large produc...
متن کاملThe antiviral compound 5-(3,4-dichlorophenyl) methylhydantoin inhibits the post-synthetic cleavages and the assembly of poliovirus in a cell-free system.
The mode of action of the antiviral drug 5-(3,4-dichlorophenyl) methylhydantoin (hydantoin) was studied in a cell-free system allowing de novo synthesis of poliovirus. This cell-free system, which is programmed with viral RNA, is suitable for the study of the late stages of poliovirus replication and, thus, for a study of antiviral compounds acting on these late stages. It was shown that, apart...
متن کاملProcessing Time Effects on Functional and Antioxidant Properties of the Quinoa Proteins Hydrolyzed with Alcalase and Pancreatin
Background and Objectives: Antioxidants are used to decrease oxidation of oils and increase shelf life of foods for centuries. Nowadays, researchers investigate for the replacement of synthetic antioxidants with antioxidants from natural sources. The purpose of this study was to investigate effects of quinoa enzyme-hydrolyzed proteins on functional and antioxidant properties of the produced pep...
متن کاملAlarm of Circulating Wild Poliovirus and Of Vaccine-Derived Poliovirus in Middle East Countries as a Potential Risk for Re-Emerging of Polio in Iran
Poliomyelitis is a highly infectious viral disease which is mainly transmitted via contaminated food and drinking water by human feces, especially in situations of poor hygiene and sanitation (1). This disease is caused by 3 types of wild poliovirus (WPV) (types 1, 2 and 3), and immunity against one type does not create immunity against other types. In our country according National Immuni...
